    ASTM E2149: Standard antimicrobial test method for assessing antimicrobial activity of immobilized agents under dynamic conditions.

    ASTM E2149 Antimicrobial Testing Overview

    The ASTM E2149 standard test method evaluates the resistance of non-leaching antimicrobial-treated specimens to microbial growth under dynamic contact conditions.

    In this method, samples are agitated in dynamic shake flasks using a wrist-action shaker to ensure optimal contact between the bacteria and the treated substrates.

    Microbial concentrations in treated and untreated samples are measured at the start (zero hours) and after specified contact times, typically at 1 or 24 hours, as determined by the sponsor.

    Organisms: Klebsiella pneumoniae, ATCC No. 4352. Other organisms like Staphylococcus aureus , E.coli may be tested upon request.

    Sample Requirements: Between 0.5 and 2.0 grams

    Contact time: 1 hr. unless a different contact time is specified

    Turn around time: 5 to 8 days

    ASTM D3273: Standard antimicrobial testing method for resistance to mold on the surface of interior coatings in an environmental chamber.

    Purpose of ASTM D3273 Test

    The ASTM D3273 test is designed to evaluate the resistance of interior coatings to fungal defacement, specifically assessing mold growth on their surfaces. This antimicrobial testing method utilizes a controlled environmental chamber to simulate severe interior conditions over a four-week period, allowing for a reproducible evaluation of paint films against mold and mildew.

    Test Panels for ASTM D3273 Antimicrobial test are prepared in triplicate . Two coats of the antimicrobial test material are applied to both faces and to all edges of the panels allowing 1 day between coats unless otherwise specified.

    ASTM E2180: Standard antimicrobial testing method for determining the activity of incorporated antimicrobial agents in polymeric or hydrophobic materials.

    Purpose of ASTM E2180 Test

    The ASTM E2180 test quantitatively evaluates the antimicrobial effectiveness of agents incorporated into or onto hydrophobic or polymeric surfaces. On such surfaces, it is challenging to achieve uniform microbial contact, which is necessary to assess antimicrobial activity. ASTM E2180 uses an inoculated agar slurry to facilitate intimate contact between microorganisms and the treated surface.

    ASTM E2180 Test Method Overview

    • An agar slurry is inoculated with a standardized culture of bacteria (e.g. Staphylococcus aureus, Pseudomonas aeruginosa) or other microbes.
    • The inoculated agar is spread onto the test material and untreated controls in triplicate.
    • After the contact time (typically 24 hours), surviving microbes are recovered by eluting the agar from the substrate into neutralizing broth.
    • Serial dilutions are plated, incubated, and colonies counted to determine percent reduction on treated vs untreated samples.
    • Exposure times can be extended up to 96 hours for more resistant microbes.

    This method can confirm antimicrobial activity, quantify differences between treated and untreated surfaces, and assess long-term durability by testing washed and unwashed samples over time.

    Turn Around Time: Eight to ten working days (if commonly tested organisms are tested.)

    Sample Requirements: 3.0 x 3.0 cm square samples of the treated and control test materials (

    Number of Samples: Since each time point requires 3 test pieces, for example for time 0 hour and 24hours= 3×2 = 6 pieces will be required per organism ordered.

    An untreated control sample is also required.

    Test Organisms: Staphylococcus aureus ,Pseudomonas aeruginosa or Klebsiella pneumoniae .Other microorganisms such as yeast or fungi may also be tested using this procedure.

    ASTM D4300: Standard antimicrobial testing method for adhesive films for their ability to resist fungal growth.

    This test method measures changes of a population of aerobic microorganisms within a specified time period when tested against antimicrobial test materials. There are multiple options to consider when conducting an ASTM E2315 time-kill testing study. These are important to define when conducting this test in order to ensure that certain variables can be distinguished and standardized. These can include:

    The ASTM E2315 Time-Kill Testing procedure allows a comparison analysis of tests done under multiple different and varying environments, circumstances, etc.

    This test’s main purpose and importance is held in that it can be used to evaluate the decrease of a microbial population of test organisms after being exposed to certain test products and materials.

    ASTM G21: Standard practice for determining resistance of synthetic polymeric materials to fungi.

    ASTM G21 Test Method Overview

    The ASTM G21 test method assesses the resistance of synthetic polymeric materials to fungal growth. This includes a range of materials such as paints, plastics, paper, cardboard, and drywall, which may be in various forms like tubes, rods, films, or sheets.

    Synthetic polymeric materials are typically resistant to fungi because they do not provide a carbon source that supports fungal growth. However, other components, such as lubricants and colorants, can promote fungal proliferation on these materials. Understanding the conditions that favor microbial growth is crucial during the production of plastic materials.

    The ASTM G21 test determines whether synthetic polymeric materials can resist fungal invasion under various environmental conditions, including different temperatures and humidity levels.

    Applications of ASTM G21 Testing

    • Assessing Antifungal Activity: Evaluates the antifungal properties of paints and coatings.
    • Evaluating Plastic Materials: Tests the antimicrobial effectiveness of PVC piping and plumbing materials.
    • Analyzing Construction Materials: Assesses the resistance of synthetic polymer-based construction materials to mold and bacteria.

     

    ASTM G21 Testing Procedure

    1. Preparation: High concentrations of spore suspensions from five fungal species are prepared.
    2. Inoculation: Nutrient Salt Agar is poured into sterile Petri dishes, and antimicrobial-treated test specimens are placed on the agar surface.
    3. Exposure: A composite spore suspension is sprayed onto the agar and test specimens.
    4. Incubation: The inoculated specimens, along with untreated positive and negative controls, are incubated at 28 to 30°C with over 90% relative humidity for 28 days.

    This method is widely used to evaluate the resistance of materials to fungal attacks, providing essential data for manufacturers to ensure product durability and performance.

    ASTM G21 Test Sample Requirements:

    Three to five pieces of 2 x 2 inch size
    The simplest specimen may be a 50 by 50-mm (2 by 2-in.) piece, a 50-mm (2-in.) diameter piece, or a piece (rod or tubing) at least 76 mm (3 in.) long cut from the material to be tested.
    Film-forming materials such as coatings may be tested in the form of films at least 50 by 25 mm (2 by 1 in.) in size.
    If the specimen is different on two sides, three specimens of each, face up and face down, will be tested upon Sponsor request.

    ASTM G21 Turn Around Time:

    Test itself is 28 days test. Turn around time for ASTM G21 test is 4 to 5 weeks.
